Abstract: The mammalian placenta – a unique and complex organ – owes much of its innovation to the co-option of ancient retroviruses. Recent research has identified syncytins and other ERV-derived genes across placental mammal lineages (e.g. primates, rodents, ruminants, carnivores, rabbits, and marsupials), each originating from independent retroviral insertions. These viral env genes mediate trophoblast cell fusion (forming the syncytiotrophoblast) and contribute to immune modulation, while ERV LTRs have been repurposed as regulatory elements in placental gene networks. For example, humans use HERV-W (Syncytin‑1) and HERV-FRD (Syncytin‑2) env proteins for trophoblast fusion pubmed.ncbi.nlm.nih.gov pubmed.ncbi.nlm.nih.gov. Mice have two distinct murine syncytins (Syncytin-A and B) pubmed.ncbi.nlm.nih.gov. Lagomorphs (rabbits) acquired syncytin-Ory1 ~12–30 MYA, mediating cell fusion via the ASCT2 receptor pubmed.ncbi.nlm.nih.gov pubmed.ncbi.nlm.nih.gov. Ruminants bear Syncytin‑Rum1 (in cattle, sheep, goats) and a younger Fematrin‑1/BERV-K1 (in bovines) mdpi.com; sheep also exploit enJSRV env for trophoblast growth. Carnivores (dogs/cats) harbor Syncytin-Car1, active for ≥70 MY in placental syncytialization link.springer.com link.springer.com. Even non-eutherians (a South American opossum) have Syncytin-Opo1pubmed.ncbi.nlm.nih.gov, showing that independent viral captures shaped marsupial trophoblasts. In each lineage, the viral origin, host species, and molecular role (fusogen vs. immunosuppressant or regulator) have been documented. For instance, human Suppressyn (ERVH48-1) antagonizes cell fusion pubmed.ncbi.nlm.nih.gov, and several env proteins retain retroviral immunosuppressive motifs (as in marsupials pubmed.ncbi.nlm.nih.gov) to modulate maternal–fetal immune tolerance. Beyond fusogens, ERV-derived elements have rewired placental gene expression: e.g. an anthropoid-specific LTR (THE1B) drives placental CRH hormone expression mdpi.com, and MER41 LTRs distribute interferon-responsive enhancers in trophoblasts mdpi.com. Recent studies continue to reveal obscure ERV contributions, from caviomorph rodents to hyenas, enriching the tapestry of placental evolution.
